Phosphatidic Acid, Lysophosphatidic Acid and Related Lipids

چکیده
Phosphatidic acid or 1,2-diacyl-sn-glycero-3-phosphate is not an abundant lipid constituent of any living organism to my knowledge, but it is extremely important as an intermediate in the biosynthesis of triacylglycerols and phospholipids and as a signalling molecule. Indeed, it is often over-estimated in tissues as it can arise by inadvertent enzymatic hydrolysis during inappropriate storage or extraction conditions during analysis. It is the simplest diacyl-glycerophospholipid, and the only one with a phosphomonoester as the head group. The molecule is acidic and carries a negative charge, i.e. it is an anionic lipid.

What makes the bioactive lipids phosphatidic acid and lysophosphatidic acid so special?
Phosphatidic acid and lysophosphatidic acid are minor but important anionic bioactive lipids involved in a number of key cellular processes, yet these molecules have a simple phosphate headgroup. To find out what is so special about these lipids, we determined the ionization behavior of phosphatidic acid (PA) and lysophosphatidic acid (LPA) in extended (flat) mixed lipid bilayers using magic an...
متن کاملQuantification of phosphatidic acid and lysophosphatidic acid by HPLC with evaporative light-scattering detection.
Phosphatidic acid (PA) and lysophosphatidic acid (LPA) are lipids that regulate cellular processes. PA stimulates kinases and may play a role in exocytosis and membrane fusion. LPA can induce cell proliferation, platelet aggregation, and microfilament formation. Due to the growing interest in these lipids, rapid purification and quantification of these lipids is desirable. We now describe a met...

Growth-inhibitory concentrations of racemic snl(3)-dodecylglycerol inhibit the incorporation of [’“C] glycerol into lipids and lipoteichoic acid of Streptococcus mutans BHT and alter the per cent composition of the glycerolipids. Increases in phosphatidic acid and diphosphatidylglycerol (at the expense of phosphatidylglycerol) contribute the most to the change in lipid composition. No cellul...
متن کاملPPARγ Networks in Cell Signaling: Update and Impact of Cyclic Phosphatidic Acid
Lysophospholipid (LPL) has long been recognized as a membrane phospholipid metabolite. Recently, however, the LPL has emerged as a candidate for diagnostic and pharmacological interest. LPLs include lysophosphatidic acid (LPA), alkyl glycerol phosphate (AGP), cyclic phosphatidic acid (cPA), and sphingosine-1-phosphate (S1P). These biologically active lipid mediators serve to promote a variety o...
متن کاملBiosynthesis of phosphatidic acid from lysophosphatidic acid and palmityl coenzyme A.
In the preceding paper evidence was presented for the biosynthesis of lysophosphatidic acid from monoglyceride and adenosine triphosphate (1). Exogenous monoglycende was found to stim.ulate the formation of phosphatidic acid as well as lysophosphatidic acid. This observation suggested that lysophosphatidic acid was being converted to phosphatidic acid and therefore was functioning as an interme...
